Research Scientist I/II, Protein Biochemistry


Werewolf Therapeutics is an exciting, newly-public, biotherapeutics company located in the heart of the Watertown, MA biotech hub, focusing on the development of novel biologic agents targeting immune regulatory pathways for the treatment of cancer.


The successful candidate will be a creative scientist, with strong problem-solving skills and demonstrated experience in protein biochemistry, antibody/therapeutic protein development, and assay development.  H/she will be responsible for protein expression, purification, and characterization efforts, with emphasis on production, biochemical assays, and cell-based assays. H/she will be required to design and execute experiments and provide data, as well as present the data to the project team and senior management.  H/she will be expected to thrive in a dynamic start-up environment.

  • Express proteins by transient transfection.
  • Utilize and optimize protein purification schemes to isolate proteins of interest.
  • Perform biochemical characterization of proteins by analytical SEC, SDS-PAGE, BLI, or other in vitro assays.
  • Develop and apply cell-based assays to characterize protein function.
  • Perform bacterial transformations and extraction of plasmid DNA.
  • Maintain mammalian cell cultures.
  • Accurately report results and methods, including recording information in an electronic lab notebook.
  • Contribute to preparation of standard operating procedures and other reports.
  • Communicate efficiently with team members and management, and present data to colleagues and management.
  • Participate in general laboratory maintenance (such as lab equipment upkeep and managing laboratory waste).
  • Stay current on all required safety training and maintain safe lab practices such as use of PPE.
  • Other duties as may be required.



  • A Bachelor’s degree in Biology, Biochemistry, or related field, as well as 4+ years of laboratory experience is required.

Experience and Requirements:  

  • A minimum of two years of prior industry experience is required.
  • Experience in recombinant protein (e.g. antibodies, cytokines, and antigens) expression, purification, and characterization in mammalian cell expression systems, including CHO and HEK293 shake flask systems.
  • Experience with bacterial transformation and DNA extraction.
  • Experience with biochemical assays, such as SPR/BLI and ELISAs.
  • Experience running SDS-PAGE and Western blots.
  • Experience with cell-based assays.
  • Strong analytical and data analysis skills are required.
  • Strong attention to detail and organizational skills.
  • Knowledge of oncology or immunology is beneficial.
  • Experience with protein engineering and construct design is beneficial.
  • Excellent problem-solving skills for developing creative solutions and for maintaining timelines.
  • Strong written and oral communication skills are a must. Demonstrated ability to work as part of a team to execute experimental plans and deliver results is also necessary.

Computer Skills: 

  • Expertise with Microsoft Excel, PowerPoint and Word, as well as graphing software (i.e GraphPad Prism) is required. Experience with PyMOL, Snapgene, and electronic lab notebook is beneficial.


  • Work is normally performed in a typical interior/office/lab work environment.
  • None or very limited physical effort required.
  • Work environment involves some exposure to hazards or physical risk, which require following basic safety precautions.
